Areas of Research Focus:
Dr. Filler's laboratory research interests are on the pathogenesis of infections caused by the fungi, Candida albicans and Aspergillus fumigatus.
C. albicans is the major fungal pathogen of humans. Dr. Filler's lab is investigating the mechanisms by which C. albicans invades and damages vascular endothelial cells during hematogenously disseminated candidiasis. In parallel studies, they are determining how this organism invades and damages epithelial cells during mucosal candidiasis. His lab has discovered the first fungal invasin, Als3, which is expressed on the surface of C. albicans hyphae and mediates invasion by binding to cadherins on the host cell surface. His group is currently working to identify additional C. albicans invasins and their cognate host receptors. Of note, Als3 is a promising vaccine target and an anti-Als3 vaccine is currently in preclinical development.
Another area of Dr. Filler's research is A. fumigatus, which is the most frequent cause of invasive mold infections in humans. The virulence mechanisms of A. fumigatus are poorly understood. Dr. Filler's lab is identifying transcription factors that govern the virulence of this organism. The target genes of these transcription factors are then identified by microarray analysis and their functions are determined by constructing and analyzing deletion mutants. These studies have determined that there is significant transcriptional rewiring between A. fumigatus and the yeasts. Transcription factors in A. fumigatus have completely different targets and govern virulence much differently compared to their orthologs in C. albicans and other yeast.
